RESUMO

Objective To study the iodine nutrition status and analyze the detection of thyroid nodules and its related influencing factors in 450 health check-up residents in Chengdu area and to provide evidence for the prevention of thyroid nodules. Methods A total of 450 residents who underwent health check-up in the Western Theater General Hospital from January 2019 to January 2022 were selected as the research subjects. The gender, age, weight and other basic conditions of the subjects were investigated, and their urinary iodine levels and occurrence of thyroid nodules were examined. Univariate and multivariate methods were used to analyze the influencing factors of occurrence of thyroid nodules. Results The overall urinary iodine level of the 450 health check-up people in Chengdu area was (96.89 -212.38) μg/L, with an average of (164.86±42.58) μg/L. The urinary iodine level of males was significantly higher than that of females (P60 years old (P60 years old , people in rural areas and people with history of diabetes mellitus in the thyroid nodule group were higher than that in the non-thyroid nodule group, and thyroid stimulating hormone (TSH), thyroglobulin antibody (TGAb) and anti-thyroid peroxidase antibody (TPOAb) were higher than those in the non-thyroid nodule group (P60 years old, history of diabetes mellitus and high levels of TGAb and TPOAb were risk factors for the occurrence of thyroid nodules (P<0.05). Conclusion The overall iodine nutrition level of 450 health check-up people in Chengdu is in the appropriate range, and the detection rate of thyroid nodules is high. It is necessary to strengthen the examination of thyroid nodules in key populations (women , history of diabetes mellitus, the elderly, etc.), and provide early detection and active intervention to prevent the occurrence and progression of thyroid nodules.

RESUMO

Objective:To assess the value of ultrasonography in the diagnosis of fetal isomerism syndrome in the first trimester. Methods:Sonographic features of 15 fetuses with isomerism syndrome diagnosed in the Maternal and Child Health Hospital of Guangxi Zhuang Autonomous Region from January 2015 to March 2020 were reviewed. Ultrasonic characteristics in the first trimester were analyzed, and the ultrasonic characteristics of early mid-trimester and pathological anatomical results were combined for comparison. Results:There were 6 cases of left isomerism syndrome (LIS) and 9 cases of right isomerism syndrome (RIS) in the 15 fetues.Increased nuchal translucency(NT) (≥3.0 mm, 6 cases), reversed A wave in ductus venosus (10 cases), and atrio-ventricular valve regurgitation (14 cases) were found during the first trimester. There were 14 cases with abnormal visceral laterality. Of the 15 fetues, 14 cases with cardiac malformations, including 6 cases of functional single ventricle, 8 cases of complete atrioventricular septal defect (CAVSD), and 12 cases with great artery abnormalities. All of the 6 LIS cases had bradycardia, 3 cases had interruption of inferior vena cava (IVC). Six cases of RIS had juxtaposition of descending aorta and IVC, and 1 case of RIS had total anomalous pulmonary vein drainage. The major structural malformations were consistent with the early mid-trimester ultrasound examination or autopsy. Karyotype and chromosomal microarray were available in 12 cases and all were normal. Conclusions:Isomerism syndrome has high positive rate of fetal aneuploidies ultrasonographic marker, especially with the atrio-ventricular valve regurgitation, but the risk of chromosome abnormality is low. Ultrasound screening for fetal cardiac structural abnormalities is beneficial to the early diagnosis of isomerism syndrom in the first trimester.

RESUMO

Cancer is a threat to human health. New treatments for cancer can significantly prolong the survival time of patients, but fail to improve the adverse reactions induced by chemoradiotherapy. Improving patient outcomes still requires the effort of cancer researchers. In recent years, the anti-tumor effects of active components from Chinese herbs have received wide attention. Kaempferol, a flavonoid mainly found in the medicinal plant Kaempferia galanga, can be used to treat obesity, cardiovascular diseases, diabetes and other diseases. It has also exhibited good efficacy in inhibiting the occurrence and development of liver cancer, colon cancer, lung cancer, ovarian cancer, and other malignant tumors. Kaempferol mainly exerts the anti-cancer effect by inducing apoptosis. Specifically, it promotes the production of intracellular reactive oxygen species (ROS) and triggers cell apoptosis through the mitochondrial pathway. Besides, it is capable of interfering with the cancer cell cycle, causing most cancer cells to arrest in the G2/M phase, and inducing cell autophagy, a programmed cell death, thus inhibiting cell migration and invasion and angiogenesis, synergistically improving chemotherapeutic drug efficacy, and reducing adverse effects. Kaempferol acts on a series of intracellular and extracellular targets to participate in the regulation of tumor cell signaling pathways, involving phosphatidylinositol 3-kinase (PI3K)/protein kinase B (Akt), epidermal growth factor receptor (EGFR), mitogen activated protein kinase (MAPK), and Wnt signaling pathways, with the PI3K/Akt signaling pathway being most significant. In addition, kaempferol also plays an important regulatory role in tumor epigenetics. This paper reviewed the anti-tumor effect and mechanism of kaempferol, aiming to provide reference for in-depth study on its prevention and treatment of tumor and the development of new anti-tumor drugs.

RESUMO

MicroRNA (miRNA) is a type of highly conserved nucleotide sequence composed of 18 to 25 nucleotides, which can specifically bind to the 3'-noncoding regions of mRNA, and then play a negative regulatory role in degrading mRNA or inhibiting translation. Long non-coding RNA (lncRNA) is a type of nucleotide sequence that exceeds 200 nucleotides in length and cannot encode proteins or can only encode protein peptides. It regulates gene expression at the levels of epigenetic, transcriptional and post-transcriptional. As an important energy storage organ, fat plays an important role in regulating the energy balance of animals, and is closely related to meat production traits such as meat production and meat quality. And the disorder of fat function can lead to hyperlipidemia, type 2 diabetes and a series of cardiovascular diseases, so the molecular regulation mechanism of animal fat deposition has attracted more attention. In recent years, more and more studies have found that miRNA and lncRNA play a crucial role in animal fat deposition. We review here the current research progresses in the role of miRNA and lncRNA in animal fat deposition, to provide theoretical guidance and new ideas for further revealing the molecular regulation mechanism of animal fat deposition.

RESUMO

Objective:To investigate the echocardiographic characteristics, pathological anatomy and genetic abnormality of congenital absent semilunar valves in first trimester. Methods:Eleven cases of congenital absent semilunar valve fetus diagnosed at 11-13 + 6 weeks of gestation in Guangxi Magernity & Child Healthcare Hospital from December 2014 to December 2018 were analyzed. The characteristics of echocardiography and the abnormal microanatomy of cardiac tissue after labor induction was compared. Results:The crown-lump length of the 11 fetus was 46-74(62.0±9.2)mm, and nuchal translucency thickness(NT) was 2.4-10.4 (6.4±2.6)mm. The NT of 10 cases were greater than 3.0 mm. Color Doppler flow imaging revealed that biphasic bidirectional flow in the aortic arch and/or pulmonary artery at the 3VT view( "to-and-fro" ) in those 11 cases, and pansystolic turbulence and pandiastolic reflux spectrum were showed on spectral Doppler. Among them, there were 10 cases of " stealing type" , including 2 cases of isolate absent aortic valves, 3 cases of absent pulmonary valves and 5 cases of absent both aortic and pulmonary valves; and all the 10 cases had secondary ultrasonic manifestations of fetal heart failure. Chromosome analysis and detection of genes showed that 4 cases with Trisomy 13 syndrome, 3 cases with Trisomy 18 syndrome, 1 case with 22q11.2 deletion, 1 case with 12q24.32q24.33 deletion and 1 case was normal. Pathological anatomy revealed enlarged heart in 8 cases, isolate absent aortic valves in 2 cases (1 case complicated with pulmonary atresia, absence of ductus arteriosus and thymus), absent pulmonary valves in 3 cases, absent both aortic and pulmonary valves in 3 cases, relics of semilunar valves in 3 cases. And 2 cases of absent pulmonary valves and 3 cases of absent both aortic and pulmonary valves with short and thick ductus arteriosus. Only 1 case was congenital absent semilunar valve in " non-stealing type" without secondary ultrasonic manifestations of fetal heart failure. Chromosome analysis and detection of genes showed 22q11.2 deletion, and there were relics of semilunar valves in the pathological anatomy. It also combined with tetralogy of Fallot and absence of ductus arteriosus. Conclusions:In first trimester, congenital absent semilunar valves are more common as " stealing type" . The echocardiographic features of congenital absent semilunar valves are the " in-out sign" of aorta arch and/or pulmonary artery and biphasic spectrum in spectral Doppler. Trisomy 13 syndrome and trisomy 18 syndrome significantly increased the risk of congenital absent semilunar valves in " stealing type" in first trimester.

RESUMO

BACKGROUND: Previous studies have observed the expression of formyl peptide receptors (FPRs) in neural stem/progenitor cells (NSPCs) and confirmed that FPRs can promote the migration of NSPCs and induce them to differentiate into neurons. FPRs ligands are present in damaged tissues, but the binding of different ligands with FPRs may lead to different and even opposite biological effects. OBJECTIVE: To investigate the effect on the differentiation of NSPCs into neurons after the binding of the ligands produced following spinal cord injury with FPRs. METHODS: Immunofluorescence staining, western blot and flow cytometry were used to analyze the expression of FPRs in NSPCs. Immunofluorescent staining with confocal microscope detection was used to analyze the effect of homogenates of the spinal cord on the differentiation of FPR1+or FPR2+NSPCs into neurons. RESULTS AND CONCLUSION: Some of NSPCs expressed FPR1 and FPR2, not only on the cell membrane, but also in the cytoplasm. The expression level of FPR1 was obviously lower than that of FPR2. The homogenate group for FPR1+or FPR2+NSPCs could produce more β-III tubulin-positive cells and fewer GFAP-positive astrocytes, and the effects could be blocked by FPR1 or FPR2 inhibitor Boc2 or WRW4. These experimental findings show that the spinal cord homogenate can induce FPR1 or FPR2 positive NSPCs to differentiate into neurons and inhibit their differentiation to astrocytes, and moreover, this effect is specific.

RESUMO

A new all-solid state electrode modified with poly ( o-aminophenol-co-o-phenylenediamine ) film was prepared by cyclic voltammetry. The electrochemical response of the electrode to various solutions in the available acidity range of 1.00 × 10-12-10.0 mol/L was studied by cyclic voltammetry. The results showed that the reduction peak potentials and the available acidity of solution showed good linear relationship in 3 stages of 1.00×10-10-0.10 mol/L, 0.10-2.00 mol/L and 2.00-10.0 mol/L respectively, and the regression equations were Epc=0.0456lgC(H+)-0.295, Epc=0.0803c(H+)-0.309, and Epc=0.0278c(H+)-0.188, respectively. There were similar responses in the solutions of H2 SO4 , HNO3 and HClO4 in the concentration range of 1.00-8.00 mol/L.The preparation method of electrode was simple, and the determination range of available acidity was as wide as 1.0 ×10-10-10.0 mol/L, which laid the foundation for the development of solid-state electrochemical sensors for determination of acidity of high acidity solution.

RESUMO

To study the effect of aqueous extracts of Yiqi Jiedu formula (YQ) on the proliferation of CNE2 cells in human nasopharyngeal carcinoma, and investigate its mechanism to provide a new theoretical basis for the clinical application of YQ. CNE2 cells were treated with different concentrations (0.125, 0.25, 0.5, 0.25 g·L⁻¹) of YQ, positive control medicine (cisplatin 4.0 mg·L⁻¹), inhibitor PD98059 (50 μmol·L⁻¹), activator isoproterenol hydrochloride (20 μmol·L⁻¹), activator isoproterenol hydrochloride (ISO)+YQ 0.5 g·L⁻¹. Then cell labeling by using real-time analyzer (RTCA) and CCK 8 method were used to detect cell proliferation activity, and the half inhibitory concentration (IC₅₀) was calculated. The cell cycle distribution was detected by fluorescence double dye flow cytometry PI staining, and Western blot method was used to detect the expression levels of related protein and MAPK/ERK signaling pathway. The results of RTCA and CCK-8 test showed that as compared with the control group, YQ group could effectively inhibit the proliferation of CNE2 cells (<0.01), with a dose and time dependence, and 48 h IC₅₀ value was 0.5 g·L⁻¹. The results of cell cycle showed that after 48 h of water extract treatment, the cell cycle was significantly changed, the proportion of G₀/G₁ was reduced, the ratio of G₂/M increased, and the cell cycle was in G₂/M period (<0.01). Western blot results showed that after 48 h treatment with different concentrations of aqueous extract, cell cycle-related proteins cyclinD1, cyclinD3 and CDK2 expression levels were down-regulated; MAPK/ERK signaling pathway related protein p-c-Raf, p-MEK, p-ERK1/2 expression level significantly lower as compared with the control group (<0.05). After adding activator and inhibitor in MAPK/ERK signaling pathway on this basis, the results showed that after adding activator ISO, cell proliferation was significantly higher than that in the Control group; the cycle related proteins cyclinD1, cyclinD3, and CDK2 expression levels were increased; at the same time, key protein p-c-Raf, p-MEK, p-ERK1/2 expression levels in the signal pathways were relatively increased. While after the addition of inhibitor PD98059, the cell proliferation was significantly lower than that in the Control group, and the expression level of corresponding protein was decreased, which was significantly different from the Control group (<0.05). So YQ could block cell cycle and inhibit the proliferation of CNE2 cells mainly by reducing the expression of MAPK/ERK signaling pathway key protein p-c-Raf, p-MEK and p-ERK1/2.

RESUMO

Objective To analyze the clinical effect of nutritional support for postoperative patients with esophageal cancer with fat emulsion and amino acids (17) and glucose (11%) injection (Calvin injection). Methods A total of 68 patients with esophageal cancer treated in the chest surgery in our hospital from December 2014 to December 2016 were selected and randomly divided into two groups with 34 cases in each group. All patients were treated with surgery for esophageal cancer. The patients in the study group were treated with Calvin injection after operation while patients in the control group were treated with conventional fluid replacement therapy. Subjective comprehensive assessment (SGA) was used to assess the nutritional status of the patients and the weight, serum total protein (TSP), serum albumin (Alb), transferrin (TF) content, urea nitrogen content in on one week, malnutrition improvement and complications of the patients in two groups were recorded and compared. Results The weight of the patients with esophageal cancer in the two groups after operation was decreased, and the weight loss of the patients in the study group was lower than that of the control group and the difference was statistically significant (P<0.01). The decrease degree of TSP, Alb and TF in the study group was lower than that in the control group, and the difference was statistically significant (P<0.05). The negative nitrogen balance of the two groups was improved, and the improvement of the negative nitrogen balance in the study group was better than that in the control group, and the difference was statistically significant (P<0.01). The postoperative malnutrition in the patients with esophageal cancer was improved. The improvement degree of malnutrition in the study group was better than that in the control group, and the difference was statistically significant (P<0.05). The incidence of complications in the study group was lower than that in the control group, and the difference was statistically significant (P<0.05). Conclusion Compared with the traditional method of postoperative fluid replacement, Calvin injection can improve the recovery of the body function of the patients with esophageal cancer after operation, improve the patient's negative nitrogen balance and nutritional status, reduce the incidence of complications, and improve the clinical prognosis. This treatment is safer and more effective and it is worth popularizing in clinical practice.

RESUMO

Objective:To investigate the distribution and drug resistance of anaerobic bacteria in the patients with oral and maxillofacial infection. Methods:Aerobic and anaerobic bacteria cultures from 61 specimens of pus from the patients with oral and maxillofacial infection in the Department of Oral and Maxillofacial Surgery,Peking University School of Stomatology were identified. The culture type was evaluated by API 20A kit and drug resistance test was performed by Etest method. The clinical data and antibacterial agents for the treatment of the 61 cases were collected, and the final outcomes were recor-ded.Results:The bacteria cultures were isolated from all the specimens, with aerobic bacteria only in 6 cases (9 .8%),anaerobic bacteria only in 7 cases (1 1 .5%),and both aerobic and anaerobic bacteria in 48 cases (78.7%).There were 55 infected cases (90.2%)with anaerobic bacteria, and 81 anaero-bic bacteria stains were isolated. The highest bacteria isolation rate of Gram positive anaerobic bacteria could be found in Peptostreptococcus,Bifidobacterium and Pemphigus propionibacterium. No cefoxitin, amoxicillin/carat acid resistant strain was detected in the above three Gram positive anaerobic bacteria. The highest bacteria isolation rate of Gram negative anaerobic bacteria could be detected in Porphy-romonas and Prevotella. No metronidazole,cefoxitin,amoxicillin/carat acid resistant strain was found in the two Gram negative anaerobic bacteria. In the study,48 patients with oral and maxillofacial infection were treated according to the results of drug resistance testing, and the clinical cure rate was 81 .3%. Conclusion:Mixed aerobic and anaerobic bacteria cultures are very common in most oral and maxillofa-cial infection patients.Anaerobic bacteria culture and drug resistance testing play an important role in clinical treatment.

RESUMO

Due to a great amount of data in clinical trials, the data cleansing needs to adopt a variety of measures, including the latest developed visual check approach. According to the different types of clinical data and the different stages in the course of clinical data management, this study reviews 8 types of visual graphics that show the relevance and trend among the data. The series of graphics can rapidly detect abnormal data, monitor clinical research in real-time, make the data management process much easier and improve the clinical trial efficiency and data quality.

RESUMO

Electronic case report forms (eCRFs) instead of the traditional paper case report forms (pCRFs) are increasingly used by investigators and sponsors of clinical research. We include a total of 14 phase III studies (8 pCRF, 6 eCRF) to compare paper and electronic data documentation both quantitatively and qualitatively in clinical studies. The result suggests that adaptions of electronic data capture (EDC) in clinical trials have the advantages in optimization of data capture process, improvement of data quality and earlier clinical decision compared to paper-based methods. Furthermore, the successful implementation of EDC requires accouplements with corresponding data management processes and reallocation of resources.

RESUMO

With the wide application of electronic data management (EDC), the data management is shifting to a new mode. In order to recognize the advantages of EDC, we choose 20 representative registered clinical trials, which involve 5 404 subjects and 321 sites. We found that EDC has many beneficial impacts on the course of clinical trial data management, including the process of data collection, data cleaning, data quality control and clinical trial decision-making. The result also provides a reference for the adoption of EDC in clinical trials.
